The three aromatic amino acids – phenylalanine, tryptophan and tyrosine – give rise to the products 3-nitro-L-tyrosine, 4-nitro-L-phenylalanine and 5-nitrotryptophan. The reaction equation for tyrosine is given by way of example. Furthermore, 5-nitrohistidine and 2-nitrohistidine can be formed from histidine. Of these two products, only 5-nitro-L-histidine is known to exist as a monohydrate. No hazardous substance data are available for either compound; they have therefore been classified in accordance with Section 6(14) of the German Regulation on the Classification, Packaging and Labelling of Hazardous Substances (GefStoffV).
Five test tubes are filled with
1) a sugar solution (0.1 g sugar in 10 ml water)
2) a salt solution (0.1 g NaCl in 10 ml water)
3) a solution of egg white powder (0.1 g egg white powder in 10 ml water)
4) cooked egg white in water (0.1 g cooked egg white in 10 ml water)
5) albumin solution (approx. 10 ml)

Add 5 ml of 12% nitric acid to each. The five test tubes are placed simultaneously in a water bath preheated to 80°C and heated there for a few minutes.
The colour changes and the time taken for them to occur should be recorded.
