Preparation of Dragendorff’s reagent

ID: 198235  |  No.: 652  |  Alfred Krause

School experiment from Year 5 onwards

Category: Evidence

Note: Transferring to your DEGINTU account for risk assessment and its documentation is recommended!
Reaction equation

BiX₃ + 4 KI => K[BiI₄] + 3 KX X = (OH)₂NO₃ X = simplified formula; the correct formula would be H₉Bi₅N₄O₂₂ = Bi₅O(OH)₉(NO₃)₄ MG = 1461.98 g/mol (Source: Gestis database)

Confirmatory reaction:
[R₃NH]X + K[BiI₄] => [R₃NH]K[BiI₄] + KX

Procedure

Main method (using tartaric acid)

Solution A – Bismuth nitrate and tartaric acid solution

Dissolve 1.0 g of tartaric acid in 4 ml of deionised water (warm gently if necessary). Then add 0.09 g of basic bismuth nitrate [bismuth(III) subnitrate; Bi₅O(OH)₉(NO₃)₄; 0.06 mmol]. Treat the suspension in an ultrasonic bath for 30 minutes at 30–40 °C (alternatively: stir for 1 hour at 40 °C).

Solution B – Potassium iodide solution

Dissolve 1.6 g of potassium iodide (KI; 9.6 mmol) in 4 ml of deionised water.

Combining and processing

Add Solution B to Solution A. To the orange-coloured mixture, add a solution of 1.7 g tartaric acid in 10 ml deionised water and stir for a further 15 minutes at room temperature. Then store the mixture in the fridge at 5–7 °C for at least 4 hours (or overnight). Filter off the white precipitate using a pleated filter and transfer the clear filtrate to a brown glass bottle.

  • Shelf life: at least 6 weeks at 5–7 °C.

Alternative method (using vinegar essence)

Stir 0.1 g of basic bismuth nitrate into 8 ml of vinegar essence (25 % solution) at 40 °C for 45 minutes. Add a solution of 1.0 g potassium iodide in 2.5 ml deionised water and stir for a further approx. 15 minutes. Filter the mixture, transfer the filtrate to a brown bottle and store at 5–7 °C.

  • Shelf life: approx. 4 weeks.

Testing the solution: Place 5 ml of water in a test tube and add approximately 0.5 ml of reagent. The solution must not reach the top of the tube; if it does, prepare a fresh batch of reagent.

Preparation of a spray reagent solution for thin-layer chromatography:
Mix 10 ml of vinegar essence (25% acetic acid) with 1 ml of Dragendorff’s reagent and pour into a suitable brown spray bottle. Store in the fridge.

Equipment & materials

50 ml Erlenmeyer flask; test tubes, magnetic stirrer, ultrasonic bath, funnel, filter paper, 25 ml brown glass bottle.
10 ml spray bottle (Tip: an empty, cleaned brown nasal spray bottle works well for this)

The restrictions on use specified for Dragendorff’s reagent are based on an automated classification in accordance with the ‘Guidelines on Safety in the Classroom’ (RiSU). These restrictions are determined solely on the basis of the GHS classification of the substances contained – irrespective of the very small quantities of substances actually used and the real-world exposure scenarios associated with a detection reaction for tertiary amines (alkaloids).

This schematic approach does not do justice to the actual, minimal hazard potential: only a few drops of the reagent are used in the detection reaction; significant absorption via the skin or respiratory tract is practically impossible if handled correctly.

In the authors’ opinion, the reagent can therefore be safely handed out to pupils in a dropper bottle for them to carry out the detection reaction independently, provided they have received appropriate instruction (risk assessment, safety goggles, drop-by-drop dosing, action to take in the event of skin contact).

⚠ Further hazards and notes
  • Unintentional release
Pupils must maintain a safe distance from the source of danger. To remove the contamination, safety goggles, disposable gloves and, where aerosols may be present, an FFP3-rated filtering half-mask must be worn. Contaminated objects or surfaces must be cleaned or wiped down with a damp cloth immediately and, where necessary, disinfected. Use cellulose paper for wiping and absorbing; this must then be autoclaved and disposed of. Detailed information on species-specific procedures can be found in the data sheets of the GESTIS biological agent database.

Substitution Substitution 1
The substitution of hazardous substances, as well as their forms of use and methods of application, has been assessed. This experiment is essential for teaching key learning content and can be carried out provided that the restrictions set out in the experimental instructions are observed and the protective measures specified therein are followed.
There are no known hazardous properties of the substances or other risks that would, in principle, preclude the experiment from being carried out by pupils or teachers. The list of substances in DGUV Information 213-098 at degintu.dguv.de has been taken into account.

The original recipe using concentrated hydrochloric acid or glacial acetic acid can be replaced with the supermarket product ‘vinegar essence’ (25 per cent).
An alternative version using tartaric acid results in a reagent with a longer shelf life.
